Moist Heat Sterilization Equipment

Moist heat sterilization technology is the most widely used given its versatility and effectiveness for heat resistant products. Depending on the type of product to be sterilized, different techniques are used:

  • Steam sterilization
  • Steam-air mixture sterilization
  • Steam-water mixture sterilization
Steam sterilization is basically used to sterilize different types of solid materials: clothing, stainless steel equipment, filters, component parts of other equipment, etc., and also for liquids in sealed or ventilated containers, etc.

Steam-air mixture and steam-water mixture sterilization autoclaves have been specially designed for the terminal sterilization of liquid solutions in sealed glass or plastic containers and the process controls the differential pressure existing between the inside of the container and the chamber.

Chemical Sterilization Equipment

Ethylene oxide is a very efficient sterilising agent. It is a bactericide, a sporicide and a viricide. It has a very favourable diffusion coefficient and can be used for thermo-sensitive or very delicate materials.

At present, sterilization with ethylene oxide in the pharmaceutical industry in general has been restricted given the risk it entails due to its high mutagenic and carcinogenic properties. GMP's specify that this type of sterilization should only be used when no safer sterilization system can be used.

Nevertheless, the advantages of sterilization with ethylene oxide are still greater than those of any other system in the hospital single-use product industry. The leading companies in this sector still rely on ethylene oxide for the terminal sterilization of syringes, probes, catheters, pace makers, etc. The complex design of these types of hospital products makes the penetrability of the sterilizing agent one of the most critical aspects. It is in this area where ethylene oxide is far superior to any of the alternatives: hydrogen peroxide, peracetic acid, etc.

Telstar promotes at Interpack the only automatic vial loading & unloading system which can be cleaned (CIP) and sterilized (SIP) in place, inside the freeze-dryer chamber

Telstar will promote at Interpack (Düsseldorf 2017, 4-10 May) the only automatic vial loading & unloading system for freeze-drying processes which can be cleaned (CIP) and sterilized (SIP) in place, inside the freeze-dryer chamber. Lyogistics Zero is a new generation automatic vial loading & unloading system for pharmaceutical freeze-dryers integrating a steam-sterilizable slider with no concealed parts which is moved by a contact-free magnetic drive mechanism. This innovative system enables that the transfer device, or ‘slider’, can remain in the chamber from where it undergoes cleaning and sterilization along with the internals of the freeze dryer.

The absence of mechanical links between the drive and the slider provides the system with unprecedented levels of precision and robustness. By incorporating a passive magnetic drive, the new system developed by Telstar eliminates many of the risks related to a lack of asepsis caused by the presence of mechanisms (belts, bands, chains and linkages) and electrical equipment (cables, batteries and motors) that are typically used in conventional and current loading/unloading systems.

Suitable for use in production processes involving hazardous products, Lyogistics Zero has been designed to be integrated within aseptic isolators requiring high level protection for the operator (OEL5), the product (ISO5) and the environment.

Telstar integrates semi-automatic Glove Leak Testing System in isolators

Telstar has integrated a new semi-automatic glove tester incorporating proprietary glove port recognition technology in the range of isolators designed by the company. Developed by the Technology Centre for Barrier Isolation Systems of Telstar in The United Kingdom, the semi-auto tester has been designed and manufactured as a prototype for single use. It has been tested prior to standardizing the product and expanding to a multi-glove tester and, from now on, future glove testers will be offered as an option to be integrated into the isolator/RABs control system.

The in-house 21 CFR compliant glove tester integrated in Telstar isolators provides the customer with a reliable, traceable means of testing gloves fitted to isolators or RABs in a working environment. The glove test parameters are adjustable to comply with the customer requirements and to compensate for different glove types and the system is rated for continuous use within the restrictions of normal life-cycle maintenance.

The system built up by Telstar has been recently implemented on an aseptic filling and closing isolator system and an aseptic vial filling & closing isolator, developed by the company for two international companies. Richard Hanson, Technical Manager at the Technology Centre of Telstar in United Kingdom commented that “we are pleased to announce that the Telstar RFID glove leak testing system has been a success in both of our recent projects following months of intensive research at our facility in the United Kingdom. We now look forward to offering this system as an option on all isolator/RABs equipment”.

Features

The glove tester consists of the glove test plug and umbilical lead fitted with quick release connections to enable one handed operation. All other components are mounted on-board the Isolator/RABs. The design is an evolution of the semiautomatic system used on previous Telstar projects but now includes RFID recognition technology to identify the glove being tested, improving traceability and reducing the scope for operator error. From a QA perspective this is a significant technological advancement as without this technology the operator could test the same glove multiple times and the system would not know. In addition with CFR

Part11 compliant software it will log up to 10 tests of each glove. The test results can be printed out for the client to keep as a hard copy.

Procedure

When the tester is placed into a glove tester it reads the glove number by means of a RFID sensor in the glove port and automatically inflates the seal. The test is then started using the start pushbutton on the tester plug. After the seal has inflated to hold the tester plug in place, the glove is then inflated to a predetermined pressure.

The pressure is held at this level for a preset time and then the pressure is reduced until it reaches the required level when the test starts. If the pressure does not achieve the required pressure in a set period the test aborts or can be aborted at any time with the stop pushbutton. Operation of the start or stop pushbutton will illuminate the indication lamp around the relevant button. 

The test is carried out for a preset length of time and the values monitored continuously. If the pressure drop is more than the allowed pressure the test fails but if the test runs the full time and the pressure drop is within the allowed tolerance then the test passes. At the end of the test the glove pressure is reduced and the seal deflates. Glove pressure is logged during the test and can be shown on the trend screens, either as live value or as historical data and can be printed or exported to a C.S.V file.

To help comply with CFR 21 Part 11 an audit trail is provided which logs all user logins, alarms and set point changes made on the glove tester and all user actions require a user to login to the system the user will automatically log out at a predetermined period. Finally, initial setup requires all tags to be allocated to a glove number in the setup screen to allow the system identify which glove is being tested.

A new generation of aseptic isolators with an innovative ionHP bio-decontamination

Telstar Pura is the latest generation of Sterility Test Isolators integrated with the innovative ionized Hydrogen Peroxide (ionHP) bio-decontamination system, which has proven to dramatically decrease cycle times.

The ionHP system is the latest leading-edge bio-decontamination technology for aseptic systems, developed to resolve some of the traditional difficulties associated with H2O2 bio-decontamination processes. Unlike conventional systems, the ionHP technology developed by Telstar increases the efficacy of the bio-decontamination process and reduces degradation of construction materials while using a very low concentration of H2O2. Reduced bio-decontamination cycle times are possible since the process efficiency is not affected by temperature and humidity and therefore there is no need to pre-condition the chamber prior to H2O2 injection.

Telstar Pura is a range of two, three and four glove Sterility Test Isolators, with the option of a transfer chamber for batch or continuous sterility testing. It is designed to avoid the risk of false results and to protect the product from both the process and externally generated factors that would compromise its quality. The Telstar Pura range is suitable for performing both Direct Inoculation and Filter Membrane sterility tests.

A new App to monitor laboratory freeze-drying process in real-time

Developed by Telstar, the new mobile application keeps track of temperature and pressure parameters in-real time, enabling remote monitoring of any process parameter or alarm

A new mobile App designed for real-time temperature and pressure parameters monitoring during the freeze-drying process for laboratory lyophilizers has been developed by Telstar. This new software has been customized for the Lyobeta and LyoBeta Mini lab freeze-dryers which are the standard and compact versions of a twin-vessel pilot-scale freeze-dryer designed for the field of experimentation and research and formulation, developing recipes for pharmaceutical freeze-drying processes.

Developed by the Telstar R+D department, the new mobile application has been created to monitor the freeze drying process in real-time, being able to constantly monitor temperature and pressure parameters. The software has also incorporated a mobile alert which warns the user of any problems, enabling the user to react accordingly. The new app for Lab freeze-dryers, designed to run on Android devices at the moment, will be developed for mobile IOS in the near future.

Taking advantage of the customer’s internal wireless network capabilities any Android-based portable device will be able to remotely connect to the equipment once the LyoBeta freeze dryer has been connected to the customer’s internal network. The current version of the app provides one-to-one equipment connection but multiuser connection capability, so the user can only be connected to a single freeze dryer but any freeze dryer can accept more than one user connected. In addition, the app provides a standard list of alarms/notifications that will pop-up during the process offering the possibility of configuring a desired alarm related to almost any process variable listed. 

LyoBeta and LyoBeta Mini

LyoBeta is a range of twin-vessel pilot-scale freeze dryers designed by Telstar for the field of experimentation, and research and formulation, developing recipes for pharmaceutical freeze-drying processes. The LyoBeta range encompasses flexibility, high performance, and small foot print required in a laboratory unit, as well advanced process control systems, management software and interface features. It is therefore considered one of the most complete and efficient pilot units on the market. In addition to the standard range, LyoBeta Mini is a new compact version which offers an external condenser with a capacity of 6 Kg located separately from the product chamber; an innovative feature which is unusual in this range of compact bench-top equipment. This model of pilot-scale freeze dryer developed by Telstar bridges the gap in the market that was hitherto addressed through the use of larger scale laboratory equipment.
